操作步驟(僅供參考):
1.配制65mM H2O2基液:本試劑盒提供的H2O2基液中的H2O2濃度約為1M。由于過(guò)氧化氫不是非常穩(wěn)定,使用前需自行測(cè)定過(guò)氧化氫的實(shí)際濃度。把濃度約為1M的H2O2基液用本試劑盒提供的CAT Assay buffer稀釋100倍,使H2O2基液中的H2O2濃度約為10mM。
2.準(zhǔn)備樣品:
a,細(xì)胞或組織樣品:取恰當(dāng)細(xì)胞或組織進(jìn)行裂解,可以采用Leagene Western及IP 細(xì)胞裂解液,如果有必要需進(jìn)行適當(dāng)勻漿,低速離心取上清,-70℃凍存,用于CAT的檢測(cè)。
b,血漿、血清和尿液樣品:血漿、血清按照常規(guī)方法制備,用生理鹽水10倍稀釋后, 可以直接用于本試劑盒的測(cè)定,尿液通常也可以直接用于測(cè)定,-70℃凍存,用于CAT的檢測(cè)。
c,全血樣品:收集適量的全血(whole blood)至一抗凝管內(nèi),顛倒混勻。取100μl全 血凍融一次,用CAT Assay buffer1000倍后進(jìn)行CAT檢測(cè)。
d,血液中的紅細(xì)胞裂解液:用抗凝管收集血液,顛倒混勻。取至少500μl全血4℃ 3000g離心5min,棄上清,沉淀用預(yù)冷的生理鹽水洗滌3次。
e,高活性樣品:如果樣品中含有較高活性的CAT,可以使用CAT Assay buffer稀釋。
f,(選做)樣品準(zhǔn)備完畢后可以用BCA蛋白濃度測(cè)定試劑盒測(cè)定蛋白濃度,以便于后續(xù) 計(jì)算單位蛋白重量組織或細(xì)胞內(nèi)的CAT含量。
3、 CAT檢測(cè):按照下表設(shè)置空白管、自身對(duì)照管、測(cè)定管,溶液應(yīng)按照順序依次加入,并 注意避免產(chǎn)生氣泡。如果樣品中的酶活性過(guò)高,可以減少樣品用量或適當(dāng)稀釋后再進(jìn)行測(cè)定。樣品的檢測(cè)能設(shè)置平行孔。
4、分光光度計(jì)檢測(cè)405nm處吸光度,分光光度計(jì)比色杯光徑0.5cm。如果沒(méi)有分光光度計(jì),亦可用酶標(biāo)儀檢測(cè),但如果有條件,盡量采用分光光度計(jì)檢測(cè)。蒸餾水調(diào)零,讀取各管吸光度值。一般應(yīng)數(shù)小時(shí)內(nèi)檢測(cè)完畢。
樣品制備:
1). 直接或稀釋使用清亮無(wú)色中性液體樣品,體積可達(dá)2.000ml。
2). 過(guò)濾混濁溶液。
3). 除去樣品中的CO 2 (測(cè)試盒說(shuō)明書(shū)過(guò)過(guò)濾)。
4 ). 測(cè)試盒說(shuō)明書(shū)過(guò)加氫氧化鉀或氫氧化鈉將酸性樣品的PH值調(diào)至8.0。
5 ). 調(diào)整酸性淺色樣品的PH值至8.0,孵育約15分鐘。
6 ). 用空白樣品做對(duì)照測(cè)定有色樣品(如有必要調(diào)整PH值至8.0)。
7 ). 用PVPP( 聚乙烯吡咯烷酮)或聚酰胺處理深色未經(jīng)稀釋或體積更大的樣品。
8 ). 壓碎、攪勻固體或半固體樣品,用水溶解提取。
9 ). 用Carrez試劑將含有蛋白質(zhì)的樣品去蛋白。
10).含脂肪的樣品用熱水提取。

產(chǎn)品介紹 OLFM3 is a 478 amino acid protein that interacts with myocilin. Myocilin is an extracellular protein that plays a key role in the actomyosin system and is responsible for controlling intraocular pressure. OLFM3 is a secreted protein that contains an olfactomedin-like (OLF) domain, an approximately 260 amino acid motif commonly found in secreted glycoproteins. OLFM3 localizes to the Golgi apparatus of the cell and is highly expressed in both eye and brain tissue. Mutations in the gene that encodes OLFM3 may cause severe glaucoma, a condition in which increased intraocular pressure within the eyeball causes loss of eye sight.
Subunit : Peripherally associated with AMPAR complex. AMPAR complex consists of an inner core made of 4 pore-forming GluA/GRIA proteins (GRIA1, GRIA2, GRIA3 and GRIA4) and 4 major auxiliary subunits arranged in a twofold symmetry. One of the two pairs of distinct binding sites is occupied either by CNIH2, CNIH3 or CACNG2, CACNG3. The other harbors CACNG2, CACNG3, CACNG4, CACNG8 or GSG1L. This inner core of AMPAR complex is complemented by outer core constituents binding directly to the GluA/GRIA proteins at sites distinct from the interaction sites of the inner core constituents.
