操作步驟(僅供參考)：
1.配制65mM H2O2基液：本試劑盒提供的H2O2基液中的H2O2濃度約為1M。由于過氧化氫不是非常穩(wěn)定，使用前需自行測定過氧化氫的實際濃度。把濃度約為1M的H2O2基液用本試劑盒提供的CAT Assay buffer稀釋100倍，使H2O2基液中的H2O2濃度約為10mM。
2.準備樣品：
a,細胞或組織樣品：取恰當(dāng)細胞或組織進行裂解，可以采用Leagene Western及IP 細胞裂解液，如果有必要需進行適當(dāng)勻漿，低速離心取上清，-70℃凍存，用于CAT的檢測。
b,血漿、血清和尿液樣品：血漿、血清按照常規(guī)方法制備，用生理鹽水10倍稀釋后， 可以直接用于本試劑盒的測定，尿液通常也可以直接用于測定，-70℃凍存，用于CAT的檢測。
c,全血樣品：收集適量的全血(whole blood)至一抗凝管內(nèi)，顛倒混勻。取100μl全 血凍融一次，用CAT Assay buffer1000倍后進行CAT檢測。
d,血液中的紅細胞裂解液：用抗凝管收集血液，顛倒混勻。取至少500μl全血4℃ 3000g離心5min，棄上清，沉淀用預(yù)冷的生理鹽水洗滌3次。
e,高活性樣品：如果樣品中含有較高活性的CAT，可以使用CAT Assay buffer稀釋。
f,(選做)樣品準備完畢后可以用BCA蛋白濃度測定試劑盒測定蛋白濃度，以便于后續(xù) 計算單位蛋白重量組織或細胞內(nèi)的CAT含量。
3、 CAT檢測：按照下表設(shè)置空白管、自身對照管、測定管，溶液應(yīng)按照順序依次加入，并 注意避免產(chǎn)生氣泡。如果樣品中的酶活性過高，可以減少樣品用量或適當(dāng)稀釋后再進行測定。樣品的檢測能設(shè)置平行孔。
4、分光光度計檢測405nm處吸光度，分光光度計比色杯光徑0.5cm。如果沒有分光光度計，亦可用酶標儀檢測，但如果有條件，盡量采用分光光度計檢測。蒸餾水調(diào)零，讀取各管吸光度值。一般應(yīng)數(shù)小時內(nèi)檢測完畢。
樣品制備：
1）. 直接或稀釋使用清亮無色中性液體樣品，體積可達2.000ml。
2）. 過濾混濁溶液。
3）. 除去樣品中的CO 2 （測試盒說明書過過濾）。
4 ）. 測試盒說明書過加氫氧化鉀或氫氧化鈉將酸性樣品的PH值調(diào)至8.0。
5 ）. 調(diào)整酸性淺色樣品的PH值至8.0，孵育約15分鐘。
6 ）. 用空白樣品做對照測定有色樣品（如有必要調(diào)整PH值至8.0）。
7 ）. 用PVPP（ 聚乙烯吡咯烷酮）或聚酰胺處理深色未經(jīng)稀釋或體積更大的樣品。
8 ）. 壓碎、攪勻固體或半固體樣品，用水溶解提取。
9 ）. 用Carrez試劑將含有蛋白質(zhì)的樣品去蛋白。
10）.含脂肪的樣品用熱水提取。

保存條件 Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

產(chǎn)品介紹 The hypothalamus acts as a major regulatory center involved in the control of feeding behavior and energy homeostasis. Several neuropeptides and proteins have been shown to be involved in the regulation of these processes. Orexin A (hypocretin 1) a 33 amino acid peptide and orexin B(hypocretin 2), a 28 amino acid peptide, are both derived from a common 130 amino acid precursor, prepro-orexin, which is encoded by a gene localized to human chromosome 17q21. Orexin A and Orexin B stimulate food consumption when administered intracerebroventricularly to rats. Orexin gene expression in the brain is highly restricted to distinct populations of neurons located in specific hypothalamic regions, including the lateral hypothalamic area (LHA), a region implicated in feeding behavior.
Function : Neuropeptides that play a significant role in the regulation of food intake and sleep-wakefulness, possibly by coordinating the complex behavioral and physiologic responses of these complementary homeostatic functions. A broader role in the homeostatic regulation of energy metabolism, autonomic function, hormonal balance and the regulation of body fluids, is also suggested. Orexin-A binds to both OX1R and OX2R with a high affinity, whereas orexin-B binds only to OX2R with a similar high affinity.
Subcellular Location : Associated with perikaryal rough endoplasmic reticulum as well as cytoplasmic large granular vesicles at synapses.
Post-translational modifications : Specific enzymatic cleavages at paired basic residues yield the different active peptides.
DISEASE : Defects in HCRT are the cause of narcolepsy type 1 (NRCLP1) [MIM:161400]. Narcolepsy is a neurological disabling sleep disorder, characterized by excessive daytime sleepiness, sleep fragmentation, symptoms of abnormal rapid-eye-movement (REM) sleep, such as cataplexy, hypnagogic hallucinations, and sleep paralysis. Cataplexy is a sudden loss of muscle tone triggered by emotions, which is the most valuable clinical feature used to diagnose narcolepsy. Human narcolepsy is primarily a sporadically occurring disorder but familial clustering has been observed. Note=Human narcolepsy is associated with a deficient orexin system. Orexins are absent and/or greatly diminished in the brain and cerebrospinal fluid (CSF) of most narcoleptic patients.
